E-mail Router Failure: Mailbox unavailable. The server response was: 5.7.1 Unable to relay.

There are two users in CRM that suddenly can no longer send e-mails via the E-mail Router. All other users can send e-mails just fine. The message in the Event Viewer is: "Mailbox unavailable.The server response was: 5.7.1 Unable to relay." Where can I begin to troubleshoot this issue?
